Working principle: Chromatography is a technique of separating mixture of substances into their components based on their composition and molecular structure. The process involves movement of sample between two phases namely stationary phase and mobile phase. The separation of various components takes place depending upon affinity of various molecules towards stationary or mobile phase. High performance liquid chromatography uses this technique and the mobile phase (solvent) is pumped through the stationary phase (column) at high pressure. This results in faster and better separation of various components.
HPLC when used at higher flow rates with bigger columns and with the combination of a fraction collector, can be used for collecting different fractions present in the sample. It becomes a fast method for fractionating a sample.
